Protective effect of five-flavor sophora flavescens enteric-coated capsules on inflammatory bowel disease and its molecular mechanism

This study aims to investigate the effect of five-flavor sophora flavescens enteric-coated capsules (FSEC) on TNF-α-induced inflammatory bowel disease and its molecular mechanism. Wistar Rats were divided into 6 groups: Normal control group (group A): normal diet, drinking water; Model B): 100 μg/L TNF-α; FSEC high-dose C): 100μg/L TNF-α + (432 mg/kg); medium-dose D): (216 low-dose E): (108 Positive F): 500 mg/kg sulfasalazine (SAZ). Animals in each intragastrically administered twice daily for 7 days. sacrificed 24 hours after last treatment colon tissues collected subsequent experiments. The results HE staining showed that colonic tissue TNF-α-fed animals appeared damage, while treated with was improved various degrees, histological characteristics basically recovered group, suggesting could be used treat damage. According ELISA immunohistochemistry, recovery structure rats different doses might related decrease TNF-α, IL-6, IL-17, TLR-4 NF-κB proteins expression. Western blotting, TNF-α-pretreated IEC-6 cells cultured medicated serum decreased expression TRIF IFN-γ proteins. These suggest has a protective ulcerative colitis (UC), mechanism may through inhibiting activation TLR-4/NF-κB signaling pathway preventing release factors.

In a randomized, double-blind controlled trial, 42 children with irritable bowel syndrome (IBS) were given pH-dependent, enteric-coated peppermint oil capsules or placebo. After 2 weeks, 75% of those receiving peppermint oil had reduced severity of pain associated with IBS. Peppermint oil may be used as a therapeutic agent during the symptomatic phase of IBS.
